Extending the Imanaka and Aiba';s (I-A) model of plasmid instability, a class of simple models have been developed to simulate the behavior of a recombinant microbial system in which Escherichia coli was employed. Here, the probability of losing plasmid-bearing cells is taken as a function of both time and dilution rate, while the substrate and temperature conditions are assumed constant. Three versions of the function are provided, which have been shown to simulate the experimental results well. One function gives the best agreement, where the time dependence function is of a second order. A ';wash out'; dilution rate from a non-conventional viewpoint has been identified. Furthermore, with some adjustments the second-order rate model has been used in batch growth simulation, which has been benchmarked against the I-A model.
