Owing to their physicochemical properties, titanium dioxide nanoparticles (TNPs) are widely used in numerous fields. However, workplace exposure remains a health concern. The brain is susceptible to exogenous insults, and in vivo studies have shown that TNPs can be detected in animal brains following their exposure. TNP accumulation in the brain may impair neuronal cells, leading to brain disorders. The molecular mechanisms underlying TNP neurotoxicity include oxidative stress, inflammatory responses, and apoptosis. DNA methylation has been implicated in neurotoxicity induced by other exogenous stimuli such as lead and bromodeoxyuridine. However, the involvement of DNA methylation in TNP neurotoxicity has not been reported to date. We therefore performed an in vitro study and found that TNPs could cause global hypomethylation in PC12 cells. Nevertheless, more in vivo and in vitro studies should be conducted to comprehensively assess the correlation between DNA methylation and TNP neurotoxicity.
